Whenever Amazon releases a newer version of Kindle app, we will see on-screen instructions to download and install it after we tap Check for Updates.
What to do if we want to manaually update the Kindle for PC app? Uninstall the current version first, and then don't forget to delete these 2 folders C:\Users%username%\AppData\Local\Amazon\Kindle
C:\Users%username%\Documents\My Kindle Content.
And then download the latest one (package file) to finish the installing and restart your PC.

Amazon also had been packing Kindle for PC desktop version (exe) and Kindle for Mac desktop version (dmg) for many years, but after the Kindle for Mac version 1.40 released, Amazon stopped providing later packages and started to guide customers directly to the App store to download the app version.

How to downgrade and upgrade the current version of Kindle for PC desktop? I personally recommend you uninstall the current app, restart your windows laptop, and then download your desired version (.exe) to finish the installing.
